Cooper and Seyfriend return as star-crossed lovers Sky and Sophie in the prequel/sequel to global hit Mamma Mia! and in one scene they share a transatlantic duet of ABBA classic One of Us.
Speaking to RTÉ Entertainment, Cooper said: "You’re professional about it but of course I hoped that out of that history me and Amanda had that it adds to it.
"We were really, really close, I know her really well and we’re still really good friends but the song is so moving anyway."
Cooper, who has also appeared in Agent Carter and The History Boys, dated Seyfried for two-and-a-half years after filming Mamma Mia! in 2008. He also had an eight-year relationship with his Preacher co-star Ruth Negga.
He added: "As you’re an actor and a performer you’re using and sourcing all sorts of feelings that you’ve experienced and gone through and Amanda was someone I’d had a relationship with at one stage who is now a really good friend but I think it would only enhance the roles."
Asked what his earliest ABBA memory is, Cooper said: "ABBA have been the soundtrack of my life since I can remember and I found a video from when I was 12 of all of use dressed up in my mother’s fur, fake fur, with tennis rackets doing all the ABBA tracks.
"There are two girls and two boys in my family and I’d imagine I was a Benny."
