Leo Cullen has made four changes to his side following last week's hammering of Racing 92, with Garry Ringrose once again captaining the side.
There's just one change to the backline, with Luke McGrath rotated into the starting side at scrum-half and Jamison Gibson-Park dropping to the bench, as Ross Byrne continues at out-half.
Ringrose and Charlie Ngatai pair off in the centre with Hugo Keenan at full-back, between wingers James Lowe and Jimmy O'Brien.
There are three changes in the pack, with Rónan Kelleher brought in at hooker, swapping in for Dan Sheehan, while Andrew Porter and Michael Ala'alatoa continue at prop.
Ross Molony (above) comes into the second row for his 150th Leinster appearance, and packs down alongside James Ryan, with Jack Conan restored to the team in the back row.
Elsewhere among the replacements, Jordan Larmour makes his return from a foot injury having been sidelined since early October.
Gloucester head coach George Skivington is seemingly prioritising the Premiership over the Champions Cup, with 13 changes to his side following last week's win against Bordeaux-Begles.
Chris Harris, Val Rapava Ruskin, Jonny May and Lewis Ludlow are among those rested, with full-back Lloyd Evans and wing Alex Hearle the only remaining starters from Round 1, as the Cherries focus on their Christmas Eve meeting with the Leicester Tigers in the Premiership.
