Swede Karlsson and American English lay on 15 under after play was suspended for the day following seven holes of their third rounds, with both players three under for the day.
The rain-hit tournament had seen round two completed early on day three but the players ran out of time and were due for an early start on the final day in another bid to catch up time.
Behind the leading duo are South African Rory Sabbatini and American Kevin Stadler, both three shots back on 12 under after eight and six holes respectively.
Spaniard Alvaro Quiros lies on 11 under along with American duo Jason Bohn and Chris Stroud.
Quiros was one of the big movers early in the third round after picking up six shots in 14 holes, birdies coming at the second hole and also the fourth, fifth, seventh, ninth and 12th.
Four Americans lie on 10 under - Spencer Levin, Justin Leonard, Scott Brown and Charles Howell III.
Levin, like Quiros, was among the birdies as she picked up shots on the first, three in a row from the third and successively on the eighth and ninth.
