This brings the total number of Northern Ireland cases to seven.
The seventh case is in a male who was a contact of a previously confirmed case. He has mild flu symptoms and is recovering at home.
The Public Health Agency is arranging a follow-up of people who were in close contact with him since he developed symptoms.
Earlier, Mr McGimpsey told the Assembly Northern Ireland should brace itself for more cases of swine flu in the coming days and weeks. There are over 650 cases in Britain.
With the World Health Organisation warning that an official pandemic is now close, Mr McGimpsey said the number of confirmed instances in the region was set to rise in the weeks ahead.
Two cases in Northern Ireland were confirmed last night. The man and woman had returned separately from Florida and New York. They are being treated at home with anti-viral drugs and are recovering well.
Mr McGimpsey said if incidents reached a certain level, local health authorities would have to look closely at how they manage the supply of anti-viral drugs. Northern Ireland has significant stockpiles of anti-virals, enough to treat 50% of the population, which is being increased to treat 80%.
As of yesterday there were confirmed cases in 73 countries with 25,952 people having contracting the virus and a total of 139 deaths.
